Custom Standard cDNA Library Construction
AGCT¡¦s innovative technology combines
optimized cDNA synthesis and ligation methods to maximize
quality of libraries.
The size of starting materials determines the
form of the library: for samples greater than 0.5 g of tissue or 1
mg total RNA, libraries can be built in unamplified procedure. In
the pBluescript II and lambda phage vectors, the library properties
are:
Primary clones: 2 X 106 cfu
Insert range: 0.5 - 3 kb
% recombinants: 90 - 95%
Turn around time:
pBluescriptII vector: 2 - 3 weeks
lambda phage vector (w/ amplification): 3 - 4
weeks
For 25 - 50 ng mRNA samples, libraries will be
built in amplified procedure in the pDNAscript vector. The library
properties are:
Primary clones: 1 X 107 cfu
Insert range: 0.5 - 2 kb
% recombinants: 95%
Turn around time: 2 - 3 weeks
